ABSTRACT

AIM: To observe the epithelial remodeling of femtosecond laser-assisted stromal lenticule addition keratoplasty combined with corneal collagen cross-linking(SLAK-CXL)in patients with progressive keratoconus, investigate the remodeling rules of corneal epithelial and influencing factors, and provide clinical data for further refractive correction.METHODS: Retrospective and observational study. A total of 28 keratoconus patients(29 eyes)who received SLAK-CXL from September 2020 to October 2021 were included. Preoperative and postoperative visual acuity, intraocular pressure(IOP), diopter, keratometry and corneal epithelial thickness(CET)were recorded. The trend of CET change was observed. The factors affecting CET were analyzed according to the thickness and depth of the lenticule.RESULTS: Flattest meridian keratometry(Kf)and steepest meridian keratometry(Ks)at 1mo postoperatively were significantly larger than those preoperatively(P<0.05). The thinnest corneal thickness(TCT)at 1, 2, 6mo and 1a postoperatively were significantly larger than preoperative(P<0.05). The CET changed with time, with the central CET showing a larger variation tendency. The CET of superior, superior nasal, nasal, superior temporal in paracentral area were thinned, the CET of superior, temporal, superior temporal in midperipheral area were thinned, while the CET of superior nasal was thickened in peripheral area at 1, 2, 6mo and 1a postoperatively. The variation of CET was not correlated with the thickness or depth of lenticule at 1a postoperatively(P>0.05).CONCLUSIONS: It is firstly found that the corneal morphology has changed after SLAK-CXL. CET decreases and then increases and then decreases again. At 1a postoperatively, the CET of the central and paracentral areas is thinner, while the CET of the midperipheral and peripheral areas is thicker. The degree of epithelial remodeling is not correlated with lenticule thickness or depth.

ABSTRACT

@# Objective To analyze the status of health literacy and its relationship with chronic diseases and self-reported health among residents in Qingdao, discuss the influence of health literacy on health status and provide the scientific basis for the development of health education strategies and measures. Methods The stratified multistage and probability proportionate to population size sampling(PPS sampling) method was adopted in this investigation. In 2017, a questionnaire survey was conducted among 16 700 permanent residents aged 15-69 from 10 districts in Qingdao. Results The overall level of health literacy status was 15.92%, the prevalence rate of chronic diseases was 19.31%, the self-reported health proportion of good, fair and poor were 81.68%, 12.12% and 1.71% among residents in Qingdao. Logistic regression analysis showed that, after adjusting for urban and rural areas, gender, age, education, income and occupation, health literacy was a protective factor for people with chronic diseases and self-evaluated health(OR=1.232,P=0.003;OR=1.159,P=0.033). Three aspects of health literacy were correlated with chronic diseases and self-reported health among people (all P<0.05). Conclusions Health literacy is positively correlated with the health status of residents. The improvement of health literacy is an important way to enhance the health status of residents.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effects of angelica polysaccharides (APS) on the proliferation of mouse skeletal muscle satellite cells (MSCs) and c-kit expression in different in vitro hematopoietic microenvironments.</p><p><b>METHODS</b>MSCs were primarily cultured. The desmin protein was examined by immunohistochemical assay five days later. The MSCs were randomly divided into 8 groups, i. e., the control group, the supernatant from cultured bone marrow stroma cells group, 100, 200, 300 microg/mL APS added in the DMEM/F12 medium experimental groups, and 100, 200, 300 microg/mL APS intervened medium groups. The effects of the proliferation activities of MSCs were detected using MTT method. The c-kit protein of the MSCs was stained by immunohistochemistry.</p><p><b>RESULTS</b>The desmin protein was positive in the isolated cultured MSCs. Results of MTT method showed the proliferation of MSCs in APS intervened medium groups was significant. The strong positive c-kit immunoreactivity existed in APS intervened medium groups. The strong positive c-kit immunoreactivity was present in the cytoplasmic of the MSCs in the DMEM/F12 medium experimental groups and the APS intervened medium groups.</p><p><b>CONCLUSIONS</b>The APS intervened MSC medium could effectively change the growth properties of MSCs, obviously promote the proliferation of MSCs and c-kit expression. The c-kit protein might play some regulative roles in the proliferation of the MSCs.</p>

ABSTRACT

Background Keratoconus is a bilateral,noninflammatory,gradually progressive corneal disorder characterized by progressive thinning and steepening of the central cornea.It is significant to investigate keratoconusrelated pathogenic gene for elaborating the pathogenesis and establishing early diagnosis standard and taking clinical measurement.Objective The aim of the study was to explore the relationship of visual system homeobox gene (VSX1) polymorphism and the risk of sporadic keratoconus.Methods This study was approved by Ethic Commission of First Hospital of Xi' an.Written informed consent was obtained from each subject prior to enrollment.A case-controlled study was conducted.One hundred and one Han nationality patients with sporadic keratoconus were included in this study.These keratoconus patients were clinically diagnosed by slit lamp examination and corneal tomography.Single nucleolide polymorphism (SNP) of VSX1 gene was assayed and classified using the MassARRAY SNP technique.Demography and relevant risk factors were collected from each subject by questionnaire.Eighty healthy volunteers served as controls.Chi-square test and Binary logistic regression were used to evaluate the difference in the distribution of allele frequency and genotype frequency and to analyze the association with keratoconus risks.Results SNP of two genes was found in the Chinese Han population (rs743018 (c.843+140 C>T) and rs6138482(R217H C>T)).There were no significant differences in the genotype frequency and allele frequency of the SNP of two genes in the keratoconus group in comparison with the normal control group (P>0.05).After adjustment by age and sex,SNP of two genes was not significantly associated with the risk of keratoconus (regression model:rs743018 (C>T) adjusted:P=0.35,OR=0.72,95% CI:0.37-1.43 ;rs6138482 (C>T) adjusted:P =0.48,OR=0.76,95% CI:0.35-1.64).Conclusions Gene polymorphisms of rs743018(c.843+140 C>T) and rs6138482(R217H C>T) in the Chinese Han population is not associated with the risk of keratoconus.Due to the racial difference in genotype and allele frequency,the role of the VSX1 gene in the pathogenesis of keratoconus still remains controversial,and further study needs to be developed.

ABSTRACT

<p><b>OBJECTIVE</b>To study the effects of Danggui Buxue Decoction on the hematopoietic reconstruction of mice transplanted by muscle satellite cells ( MSCs).</p><p><b>METHODS</b>MSCs were procured from newly born male mice of the homologous series. The female Kunming receptor mice irradiated with 8Gy137Cs-gamma ray were randomly divided into six groups, i. e., the blank control group, the transplanted MSC group, four groups intervened by different doses of Danggui Buxue Decoction after transplanted MSC (gastrogavage by one, three, five, and ten times of clinically equivalent dose for seven days, as the DGBX 1 group, the DGBX 2 group, the DGBX 3 group, and the DGBX 4 group). After transplantation changes of splenic colony forming unit spleen (CFU-S), white blood cells (WBC), hemoglobin (Hb), and platelet (PLT) in the peripheral blood were observed in pos-transplanted 1-, 2-, and 3-week receptor mice. The 3-week survival rate was calculated. The source of hematopoietic reconstruction was identified using PCR.</p><p><b>RESULTS</b>The desmin protein was positive in the cultured MSCs. WBC of each transplanted MSC group obviously increased at the 2nd week (P<0.05). Hb obviously increased in the DGBX 2 group, the DGBX 3 group, and the DGBX 4 group (P<0.05). WBC and Hb obviously increased in the DGBX 3 group and the DGBX 4 group when compared with the transplanted MSC group at the 3rd week (P<0.05). The recovery of PLT was significant in the DGBX 4 group (P<0.05). Compared with the blank control group at the 2nd week, CFU-S obviously increased in the DGBX 3 group and the DGBX 4 group (P<0.05). PCR results of Y chromosome in the survived transplant mice indicated that the hematopoietic cells of reconstruction female receptor mice were originated from male donors.</p><p><b>CONCLUSION</b>The hematopoiesis of mice transplanted by muscle satellite cells could be constructed after intervention of Danggui Buxue Decoction.</p>

ABSTRACT

Objective:The recombinant human retinal pigment epithelium-derived factor(PEDF)protein to be obtained and the angiogenesis of the rPEDF to be identified.Methods: PEDF gene gene was amplified by PCR and cloned into pET32a,rPEDF protein was expressed in E.coli BL21 and confirmed by SDS-PAGE and Western blot.The rPEDF was purified by Ni-NTA on denature condition.The concentration of the rPEDF was determined by Bradford method.The angiogenesis of the rPEDF was determined by chick chorioallantoic membrane(CAM) method.Results: The expression plasmid pET32a-PEDF was constructed successfully.The rPEDF was expressed with stable efficiency in E.coli BL21.The results of the CAM experiment showed that the rPEDF had notable angiogenesis effect in the concentration 0.4、0.04 ng/ml,but had no effect in 4 ng/ml.Conclusion:The PEDF gene was cloned and expressed efficiently,the angiogenesis of the rPEDF to be identified and the activity was worked in certain range.The results can facilitate studying its function and spreading its application.
